Myeloproliferative disease

Chronic myelocytic leukemia
- Morphology
   - All line (myeloblasts, lymphoblasts, erythroblasts)
   - From all stage of maturity: blasts until mature granulocytes (or eosinophils or basophils), but we only count the stage of neutrophils

- Molecular, bcr/abl
- follow-up bcr/abl gene
  - bcr ratio to abl (house keeping gene).
  - major response: bcr/abl < 0.1%
  - major molecular response:
    - 4: no copy with abl >10.000 or < 0.01%
    - 4.5: no copy with abl >32.000 or < 0.0032%
    - 5: no copy with abl > 100.000 or <0.001%.
    - these copy's are calculated using normalization curve (curve where the number of copies are known)

Myelofibrosis
- Increase production of fibrosis
- Morphology:
   - teardrop
   - plus immature granulocytes and erythroblasts (due to extramedullar production which take over from fibrotic bone marrow)

One lineage myeloproliferative
- Essential thrombocytosis
- Polycythemia vera
