Prepare a slide smear (one drop of bacteria suspension --> let it dry --> fixation using methanol --> stain with crystal violet --> wash with water --> fixed the colour with iodium (lugol)--> wash with water --> decolorize using alcohol acetone --> wash with water --> do counter stain with saffran red --> wash.
